Ibuprofen (Brufen, Caldolor)

Mechanism of action

NSAID - Inhibits cyclooxygenase 1 and 2 which results in block of production of prostaglandins, normally involved in the mechanism of inflammation and pain signalling.

Pharmacokinetics

Onset of action: 30 minutes

Duration of action: 4–6 hours

Half-life of elimination: 2 hours 

Metabolism: Hepatic 

Elimination: Kidneys

Directions for administration

Infusion of ibuprofen should be administered over at least 30 minutes.

Medicinal forms

Pills – 200 mg, 400 mg

Solution for injection/infusion (100 mg/ml)

  • 1 vial = 8 ml (800 mg)

Side-effects

Bleeding (inhibition of platelet aggregation)

Abdominal pain, nausea, vomiting

Ulcers

Nephrotoxicity

Hyperkalaemia

Proteinuria

Increased risk of thrombotic events

Fluid retention, oedema

Hypertension

Thrombocytopaenia, neutropenia, aplastic anaemia

Rash, pruritus

Asthma
